What are Endotoxins and Pyrogens?

Endotoxins are biological molecules released from the outer membrane of certain bacteria. They may remain in water even after bacteria are reduced or killed. Pyrogens are substances that can cause fever or biological reactions, and endotoxins are one of the most important types of pyrogens in laboratory water applications.


Endotoxins are not living organisms. Therefore, technologies that kill bacteria do not necessarily remove endotoxins.

Where do Inorganic Ions come from?

Endotoxins may come from:

  • Gram-negative bacteria
  • Biofilm inside water systems
  • Contaminated storage tanks
  • Stagnant distribution loops
  • Poorly maintained filters
  • Microbial growth in cartridges
  • Feed water contamination
  • Dead bacterial cells


If microbial growth is not controlled, endotoxin levels may increase over time.

How do Endotoxins affect laboratory water?

Endotoxins can interfere with biological and clinical applications. They are especially important where water comes into contact with cells, biological reagents or sensitive clinical workflows.

Endotoxin contamination can cause:

  • Cell culture stress
  • Unreliable biological results
  • Interference in molecular biology
  • Problems in IVF-related work
  • Clinical application concerns
  • Reduced confidence in biological testing

 

Because endotoxins are not necessarily removed by standard ion exchange or UV disinfection, specific barriers may be required.

How are Endotoxins controlled?

Endotoxins are controlled using:

  • Ultrafiltration
  • Proper microbial control
  • Low-dead-leg system design
  • Continuous recirculation
  • Regular sanitization
  • Appropriate final filters
  • Avoiding biofilm formation
  • Monitoring where required

 
For endotoxin-sensitive applications, ultrafiltration near the point of use is often important.
